Abstract

Mountain Ciremai as in situ conservation, reserve species of bamboo were used for variously of daily needs to handy craft. As the land changes and the increasing a need for bamboo by society, the existence of natural bamboo threatened that an inventory of natural bamboo species needs to be done. This study aims to list the species diversity and abundance of bamboo in Mountain of Ciremai West Java. Sampling is done using qualitative descriptive method which includes exploration, sampling, morphological observation and identification of the type of bamboo and measurement of environmental factors. Data obtained were then analyzed using the Shannon-Wiener diversity index to find out the diversity index species of bamboo in the area Mountain of Ciremai West Java. Based on data collection in the field, found 10 bamboo species namely G. apus J.A & J. H. (shultes) Kurz, S. Silicatum Widjaja, G. pseudoarundnacea (Stard) Widjaja, B. Lako Widjaja, B. blumeana J. A & J. H, S.Brachyladum Kurz, G. AtterHassk. Kurz, D. asper (Schult) Backer ex Heyne, D. aspervar. Thai, B. vulgaris Schard. ex J.C. The type most often found is G. apus J.A & J. H. (Shcultes) Kurz, while the highest abundance was found in D. asper (Schult) Backer ex Heyne 2,500 clump. Based on the calculation of diversity indices in the area Mountain of Ciremai Kuningan relatively low level H '= 0.902164 where H' <1.5. S.brachyladum is belongs to rare species.

Abstract

Fejervarya limnocharis are amphibians which generally can be found in agriculture area. The high intensity of pesticide application in agricultural area can lead to alteration of physiological aspects of organism inhabiting that area. Quantitative examination of hematology aspect can be conducted to evaluate physiological condition of species. This research was aimed to find out about hematology profile of F. limnocharis living in agriculture area using pesticide in West Sumatera. Thus, examinations of erythrocytes, leukocytes, hemoglobin concentrations, and hematocrit values collected from F. limnocharis living in three different areas have been conducted. The data gained have been compared with normal blood value of various amphibians species from previous research. The result acquired respectively for three locations show that total of erythrocytes were in normal range, which were 1,616 x 106/mm3; 1,539 x 106/mm3; and 1,712 x 106/mm3; hemoglobin concentration were lower than normal range, which constituted 4,06 g/dL; 4 g/dL; and 4,18 g/dL; hematocrit values have normal range, which made up 40,06 %; 40,14 %; and 45,7 %; total of leukocytes were higher than normal range, which were 27,450/mm3; 49,934/mm3; and 22,040/mm3. Usage of pesticide in agriculture area causes alteration of some hematological aspect.Keywords: Fejervarya limnocharis, hematology profile, pesticide, West Sumatera

Abstract

Tomato has three lycopene gene, namely lycopene beta cyclase (Lcy-b), lycopene beta-cyclase kromoplas (Cyc-b) and lycopene epsilon cyclase (Lcy-e). Lcy-b gene can be used as determinant of phylogenetic relationship between tomato and chili. This research aims to know phylogenetic relationship among tomatoes cultivar, namely Betavila F1, Fortuna F1 and Tymoti F1, then phylogenetic relationship between these tomatoes cultivar and tomatoes sequences Kristin KC140137.1, Darsirius KC140135.1, Pennellii XM 015217853.1, Villosum KP313876.1 and outgroup Capsicum annuum GU085266.1. Besides that, this research also aims to know the difference of nucleotide among tomatoes cultivar and the others tomato sequences and the outgroup Capsicum annuum based of alignment result. PCR (Polymerase Chain Reaction) method and NCBI primer design used to isolate Lcy-b gene. Acquired sequences were analyzed by Mega 6 and Clustal X softwares. The result of phylogenetic relationship among tomatoes cultivar showed Betavila F1 cultivar closely related to Tymoti F1 cultivar and Fortuna F1 cultivar closely related to Tymoti F1 cultivar. Phylogenetic relationship between tomatoes cultivar and the other tomatoes sequences showed Fortuna F1 cultivar closely related to Darsirius cultivar and varieties Pennellii, Betavila F1 cultivar distantly related to the other tomatoes sequences, and Tymoti F1 cultivar closely related to Kristin cultivar. The difference of nucleotide were found at Pennellii tomato that is 1 nucleotide with transition mutation, found at Villosum tomato that are 12 nucleotides with transition and transversion mutation, and found at Capsicum annuum that are 16 nucleotides with transition and transversion mutation. Phylogenetic relationship using Lcy-b gene expected to be used as a transgenic strategy to modify carotenoid content that improves tomatoes nutrition

Abstract

The research aims to study the effect of rice storage period against the growth of Sitophilus oryzae population and weight loss characteristics of rice.  The rice storage period consists of five levels: rice storage for 40, 60, 80, 100, and 120 days.  The results showed that rice storage period affects the growth of S. oryzae population, weight loss characteristics of rice, and the percentage of rice powder. The 120-day storage period may increase S. oryzae populations, weight loss characteristics of rice, and the percentage of rice powder. The relationship between the storage period and the growth of S. oryzae population, weight loss characteristics of rice, and the percentage of rice powder increased linearly. There was a positive and very significant correlation between the rice storage period and the imago of S. oryzae population (r = 0.996**; P <0.01), weight loss percentage (r = 0.990**; P <0.01), perforated grain percentage (R = 0.994**; P <0.01), and the percentage of rice powder (r = 0.971**; P <0.01).

Abstract

Fortification effects of beef cattle feces with broiler chicken feces on dry matter of elephant grass was carried in the Industrial Area of Superior manure Compost, village district of Mipiran, Padamara, Purbalingga, and Experimental Farm, Faculty of Animal Science of Jenderal Soedirman University and Soil Science Laboratory of the Faculty of Agriculture, Sebelas Maret University Surakarta. This study was aim to determine the effect of commercial broiler chicken feces as the substitute for feces of beef cattle from the aspect of organic matter content and to determine the effect of fertilizer dry matter productions of elephant grass. The materials used were the feces of beef cattle as much as 930 kg, the commercial broiler chicken feces, 270 kg and 2.4 lt EM4. This research method used Completely Randomized Design (CRD) with four treatments (R), namely: R0: 100% beef cattle feces (50 kg), R1: 85% of beef cattle feces (42,5 kg) of commercial broiler chicken feces + 15% (7,5 kg), R2: 70% of beef cattle feces (35 kg) of commercial broiler chicken feces + 30% (15 kg), R3: 55% of beef cattle feces (27,5 kg) of commercial broiler chicken feces + 45 ( 22,5 kg) with six replications.The results showed that the averages of organic matter content of R0, R1, R2, and R3 were 43,071%, 34,075%, 27,815%, and 36,098% respectively. The result of analysis of variance showed that the averages of organic matter that did not use commercial broiler chicken feces (R0) was higher than using commercial broiler chicken feces.The results of analysis showed that administration of compost on the grass did not affect significantly (P>0,05) on fresh production of elephant grass, presumably because it had not decomposed completely. Compost nutrient content was low, therefore, was not sufficient to provide additional nutrients needed by the plants

Abstract

Infectious diseases are diseases caused by pathogenic bacteria that enter the body, multiply and cause disease. One cause of the disease is a bacterial infection of Staphylococcus aureus. Treatment of infectious diseases is usually done by giving antibiotic but some types of bacteria become resistant. Utilization of plants as traditional medicine is growing rapidly and many alternative medicine used by some communities, one of which is cabbage (Brasicca oleracea var. capitata f. alba) containing antibacterial compounds such as flavonoids, saponins, polyphenols, tannins and sulfur. Such compounds capable of inhibiting the growth of bacteria by inhibiting bacterial cell wall synthesis, protein synthesis, and forming complex compounds against extracellular proteins that interfere with bacterial cell membrane integrity. This research is an experimental research with the aim to determine the inhibitory ethanol extract of cabbage (Brasicca oleracea var. capitata f. alba) against Staphylococcus aureus in in vitro. Antibacterial test method used is that pitting diffusion method. Ethanol extract of cabbage (Brasicca oleracea var. capitata f. alba) can inhibit the growth of Staphylococcus aureus on the mark with the formation of a clear zone around the wells which starts from a concentration of 20%.

Abstract

The research conducted in Enrekang, South Sulawesi aims to obtain the isolates of lactic acid bacteria from dangke and identify the lactic acid bacteria that have potential as a probiotic. Identification included physical and microbiological test. The physical tests include pH and total titrated acid test, while Microbiological testing include morphological observation of bacterial colonies, gram staining and catalase test. The results showed 2 isolates with characteristics of lactic acid bacteria, positive gram, rounded and shaped. Catalase test on all isolates showed a negative result, no gas bubbles when reacted with H2O2. Thus it was concluded that all isolates shows the characteristics of lactic acid bacteria.

Abstract

This research aimed to analyze microbiological characteristics of kombucha from various types of tea processing which tasty to drink and gave benefits to examine some types of diseases. This research used an experimental method. The kombucha microbiology of old and new colonies were analyzed quantitatively descriptive by Levene homogeneity test, Kolmogorov-Smirnov normality test, and Shapiro-Wilk test. It was continued by Kruskal-Wallis and Mann-Whitney test. The results of data analysis were the average thickness, wet weight and diameter by using Kruskal-Wallis test that the provision of various types of processed tea was significantly different with the wet weight (0.222 < 0.05) but it was not significantly different to the thickness (0.170 > 0.05) and diameter (0.637 > 0.05) the old kombucha colony. Furthermore, Mann-Whitney test obtained was significant (p-value) > 0.05. It was showed that it was not significantly different to the average data wet weight of old kombucha colony. In addition, It was significantly different to various type of tea processing to the thickness (0.015 < 0.05) fresh weight (0.017 < 0.05), diameter (0.046 < 0.05), and the sheet (0.042 < 0.05) new kombucha colony. Furthermore, Mann-Whitney test obtained was significant (p-value) > 0.05. It means that it was not significantly effective to the average data of thickness, wet weight, diameter, sheet new kombucha colonies. The best treatment was administrating of green tea (P3) in the old and new kombucha colonies were significantly different from other treatments.

Abstract

Hypercholesterolemia defines as the increasing of cholesterol serum that more than the normal limit. WHO (2011) on 2008 at Indonesian said that hypercholesterolemia prevalence of woman approach about 37,2% and for man about 32,8%. Tomato contain lycopene is one powerful antioxidant and can manage cholesterol metabolism with blocked the HMG-KoA enzyme reductase (enzyme for cholesterol synthesis in hepar). This study aims to determine the effect of tomato juice on the cholesterol levels in the blood of hypercholesterolemic rats and to find the effective dosis of tomato juice to decrease the cholesterol of hypercholesterolemic rats. The object of research covering 20 wistar rats (Rattus norvegicus) male 2-3 months old with 50-150g weight. The rats were divided into five groups and four replications: normal (K-), high-fat diet (K+), high-fat diet+tomato juice 200mg/100gBB (P1), high-fat diet+tomato juice 400mg/100gBB (P2), high-fat diet+tomato juice 800mg/100gBB (P3). Tomato juice was given orally for one month every day. At the end of the treatment of rats was dissected to take blood from the heart. Data were analyzed statistically with one-way ANAVA with α=0,05. The results showed that the tomato juice affects on the cholesterol levels of blood serum of hypercholesterolemic rats and the effective dose of tomato juice used in this study was 800mg/100gBB.

Abstract

Kayu Jawa (Lannea cormendalica (Houtt.) Merr.) is native Indonesia plant, which became a typical plant for the treatment of wounds, dysentery, and other infectious diseases in some regions in Indonesia. An effort to explore of active compounds in medicinal plants in Indonesia, had tested the antimicrobial activity of the fraction Kayu Jawa cortex (Lannea coromandelica (Houtt.) Merr). This study aims to determine the polarity of compounds which can provide the best inhibitory activity against Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Salmonella thypi, Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus mutans, Vibrio sp. and Candida albicans. Extraction of Kayu Jawa cortex done by maceration method with solvent n-hexane and methanol. Antimicrobial activity for total extracts was tested with TLC-Autobiographical and fractions of extract with best antimicrobial activity were tested by agar diffusion method. Fractionation conducted with Vacum-Liquid Chromatography (KCV) method with conditioned eluent from a solvent mixture of ethyl acetate:methanol and n-hexane:ethyl acetate. Determination of the class of active compounds done using a spray reagent.The results showed that the methanol extract and n-hexane extract inhibited the growth of bacteria Escherichia coli, Pseudomonas aeruginosa and Salmonella thypi. The fraction I of methanol extract at Rf 0.64 and fraction I extract n-hexane at Rf 0.02 showed antibacterial effectiveness for Escherichia coli, Salmonella thypi and showed strong inhibition at concentrations 500-750 ppm against Pseudomonas aeruginosa. The chemical components of the Kayu Jawa cortex (Lannea coromandelica (Houtt.) Merr.), which acts as an antimicrobial included in the class of flavonoids.
